چکیده

Abstract Introduction Canine leptospirosis has always been a differential diagnosis in dogs presenting with clinical signs and blood profiles associated kidney and/or liver disease. The conventional polymerase chain reaction (PCR) provides diagnoses, but real-time PCR-based tests provide earlier confirmation determine the severity of infection, especially acute stage, allowing early detection for immediate treatment decisions. To our knowledge, PCR not routinely adopted investigation Malaysia. This study evaluated TaqMan (qPCR) assays diagnosing compared their applicability to samples from disease against reference. Material Methods qPCR were validated using existing leptospiral isolates. Whole urine analysed PCR, LipL32(1) LipL32(2) qPCRs microscopic agglutination test. sensitivity specificity determined. Results assay had more diagnostic value than assay. Further evaluation this revealed that it could detect as low five DNA copies per high tested strains. No cross-amplification was observed other organisms. Analysing samples, 100.0% >75.0% specificity. Conclusion is sensitive, specific potential be applied future studies.

Development of TaqMan Duplex Real-time PCR for Simultaneous Detection of Chlamydia Trachomatis and Mycoplasma Genitalium

Background and Objective: Sexually infections transmitted by bacteria are one of thetherapeutic and social problemsworldwide. The Real-time PCR assay is one of the most sensitive diagnostic and screening methods for these infections. The purpose of this study wassimultaneous detection of Chlamydia trachomatis and Mycoplasma genitaliumusing the TaqMan duplex real-time polymerase chain reaction. ...

Detection and differentiation of Cryptosporidium spp. in human clinical samples by use of real-time PCR.

Real-time PCR has the potential to streamline detection and identification of Cryptosporidium spp. in human clinical samples. In the present article, we report the first such assay to allow not only detection and differentiation of the most common human pathogens, Cryptosporidium hominis and Cryptosporidium parvum, but also simultaneous amplification of a region of the small subunit (SSU) rRNA ...

Simultaneous detection and differentiation of pathogenic and nonpathogenic Leptospira spp. by multiplex real-time PCR (TaqMan) assay.

Leptospirosis, caused by pathogenic Leptospira, is one of the most important zoonoses in the world. Several molecular techniques have been developed for detection and differentiation between pathogenic and saprophytic Leptospira spp. The aim of this study was to develop a rapid and simple assay for specific detection and differentiation of pathogenic Leptospira spp. by multiplex real-time PCR (...

Development and validation of a modified TaqMan based real-time PCR assay targeting the lipl32 gene for detection of pathogenic Leptospira in canine urine samples.

A modified TaqMan real-time polymerase chain reaction targeting a 138bp fragment within the lipl32 gene was developed to identify exclusively pathogenic Leptospira spp. in dog urine samples. Thirty-five samples from dogs with suspected clinical leptospirosis and 116 samples from apparently healthy dogs were tested for presence of leptospiral DNA using the TaqMan-based assay. The results were co...

A quantitative PCR (TaqMan) assay for pathogenic Leptospira spp

BACKGROUND Leptospirosis is an emerging infectious disease. The differential diagnosis of leptospirosis is difficult due to the varied and often "flu like" symptoms which may result in a missed or delayed diagnosis. There are over 230 known serovars in the genus Leptospira. Confirmatory serological diagnosis of leptospirosis is usually made using the microscopic agglutination test (MAT) which r...
